Rabbit monoclonal antibodies for neutralization and activation/proliferation

Neutralization

Neutralizing a specific target of protein with Bioactive antibodies can give valuable insight into a protein's biological activities. Additionally, neutralization can be utilized to assist in measuring the biological activity of samples containing several bioactive proteins.

Blocking ligand-receptor interaction – Approximately 0.6 ug/ml of this antibody blocks 50% of the binding of 0.5 ug/ml of hVEGF165 to immobilized rhKDR (50 μl of a 5 μg/ml solution was coated on a 96-well plate) in an ELISA assay. Inhibition of HUVEC cell proliferation by anti-human VEGF antibody (cat# 4408-1) -To measure the ability of the antibody to neutralize the bioactivity of hVEGF on HUVEC cells, 0.4 ng/ml of rhVEGF was incubated with various concentrations of the antibody for 1.5 hour at 37°C in a 96-well plate before 1 x104 cells of HUVEC cells were added per well. The assay mixture in a total volume of 200 μl, containing antibody at the concentrations indicated, was incubated at 37°C for 72 hours. Then 20 ul alamar blue was added and incubated for a suitable time at 37°C. The plate was read using a fluorescence system (excitation at 530 nm and emission at 590 nm). The IC50 of this lot of antibody was approximately 4-10 ng/ml.




Activation/Proliferation

For activation/proliferation, bioactive antibodies can be used to induce biological activities in samples by acting as the inducing agents.
