Description

Akt1/PKBa is a Ser/Thr protein kinase whose kinase activity plays a key role in various cellular functions, including apoptosis, glycogen synthesis, and cell growth. Akt1/PKBa is activated by various growth and survival factors (1). Phosphorylation on Thr308, Ser473 and Tyr474 is required for full activity. Akt1/PKBa inhibits apoptosis by phosphorylating and inactivating several targets, including caspase-9 (2). Akt1/PKBa regulates glycogen synthesis by phosphorylating and inactivating glycogen synthase kinase-3a and b (3, 4). Three mammalian Akt1/PKBa isoforms have been identified. This kit detects human Akt1 and has no cross-reactivity with mouse and rat Akt1. This assay employs the sandwich enzyme immunoassay technique that detects human Akt1. Rabbit anti-Akt1 monoclonal antibody has been pre-coated onto a microplate. Cell lysate is pipetted into the wells. Following extensive washing, mouse anti-human Akt1 monoclonal antibody reagent is added. Following a wash to remove any unbound antibody reagent, goat anti-mouse IgG-HRP is added to the well. After washing away the unbound goat anti-mouse IgG-HRP, a substrate solution is added to the wells to develop color. The magnitude of the absorbance for this developed color is proportional to the amount of human Akt1 protein.

This kit is not intended to be a quantitative ELISA kit.
