ELISA Kit Technology Principals


1. Sandwich ELISA Assay

This assay employs the sandwich enzyme immunoassay technique that detects endogenous levels of histone H2B when acetylated at Lys5 (Fig 1). Rabbit monoclonal antibody has been precoated onto a microplate (Step 1). Cell lysate is pipetted into the wells (Step 2). Following extensive washing, biotinylated rabbit detection monoclonal antibody reagent is added (Step 3). Following a wash to remove any unbound antibody reagent, streptavidin-HRP is added to the well (Step 4). After washing away the unbound streptavidin- HRP, a substrate solution is added to the wells to develop color (Step 5). The magnitude of the absorbance for this developed color is proportional to the amount of endogenous protein.


2. Competition Assay (Anti-PEG ELISA Kit only)

Polyethylene glycol (PEG) is an O-CH2-CH2 polymer, which is water-soluble, nontoxic, nonantigenic, and biocompatible. Covalent conjugation of PEG to therapeutic proteins increases their in vivo stability by protecting the protein from degradation, masking its immunogenic sites and reducing clearance (1). Typically, PEGylation uses nonspecific reactions with nucleophilic residues and produces mixtures of PEGylated positional isomers (2). Qualitative and quantitative analysis of PEGylated molecules is important for both drug development and clinical applications. Therefore, this kit is developed to determine levels of PEGylated molecules in samples such as serum via ELISA. more info

The PEG ELISA kit operates on the basis of competition between the enzyme (HRP) conjugated PEG and PEG labeled molecules for a limited number of binding sites on the surface of 96-wells coated with anti-PEG antibody. The extent of color development resulted from interaction between HRP and substrate TMB is inversely proportional to the amount of PEGylated molecules in the sample (Figure 2.).

• Users must provide and PEGylate their molecules of interest
• Use of kit requires the end user to have at least 250ng of PEGylated compound of interest to use as reference
• PEG-BSA is included as a reference standard only
